MT-1 (Melanotan I, afamelanotide) is a linear alpha-MSH analog (CAS 75921-69-6) with a Nle4, D-Phe7 substitution pattern that enhances MC1R binding affinity and confers DPP-IV resistance relative to native alpha-MSH. It is the reference agonist for MC1R pharmacology studies in melanocyte culture models and is used to probe eumelanin synthesis pathways via the cAMP-MITF-MC1R cascade. Compared to Melanotan 2, its linear structure and receptor selectivity profile make it a distinct tool for dissecting individual melanocortin receptor subtypes.
